1-A-N is an ionizable cationic lipid (pKa = 6.8) that has been used in the generation of lipid nanoparticles (LNPs) for siRNA delivery in vivo.[1] LNPs containing 1-A-N and encapsulating siRNA targeting the gene encoding CD45 reduce its gene expression in splenic T cells in mice.
