Amylin is a 37-residue peptide hormone secreted by pancreatic β-cells that reduces food intake, modifies glycogen synthesis, slows gastric emptying, and increases satiety. It binds to heteromeric complexes of human calcitonin receptor 2 (CTR2) with receptor activity-modifying protein (RAMP) 1 or 3 (IC50s = 7.86 and 6.35 nM, respectively). It also binds to mouse α-thyroid-stimulating hormone thyrotroph (α-TSH) cells (IC50 = 10.5 nM) and rat nucleus accumbens membrane preparations (Ki = 0.024 nM). In vitro, amylin (100 nM) inhibits insulin-induced, but not basal, glycogen synthesis in hepatocytes isolated from fasted rats. It also inhibits insulin-induced glycogen synthesis ex vivo in isolated soleus muscle from fasted rats. In vivo, amylin delays gastric emptying in rats (ED50 = 69 pmol) and decreases food intake in fasted mice (ED50 = 110 μg/kg). Unlike human amylin , rat amylin does not form fibrils and is not cytotoxic.
