SYTO9 Gren fluorescent Nucleic Acid Stain (SVTO9 Gren fluorescent Nucleic Acid Stain) is an excellent counterstain for nuclei and chromosomes and is highly permeable to cell membranes. SYTO 9 binds DNA (and RNA) with high affinity. Once bound, it shows a significantly enhanced fluorescence signal. It is excited by blue light (maximum emission wavelength 485nm) and emits green fluorescence (maximum excitation is 498nm (DNA) and 501nm (RNA) respectively). . Because SYTO 9 can penetrate well into prokaryotic and eukaryotic cell membranes, it is commonly used as a nuclear counterstain (especially bacterial cells). It is often used in combination with dead cell nuclear counterstains (such as propidium iodide PI). Live cell/dead bacterial staining.
What is more noteworthy is that SYTO 9, as a sensitive DNA-binding dye, exhibits many excellent characteristics in RT-PCR, including: producing highly reproducible DNA melting curves under a wide range of dye concentrations, extremely low PCR inhibition rate, high signal-to-noise ratio and high sensitivity make it an ideal SYBR Green I alternative dye for RT-PCR, DNA melting curve analysis, and loop-mediated isothermal amplification (LAMP) experiments [3- 5].
