Cbl-b-IN-2 (Example 8) is an orally effective bioactive compound that inhibits the E3 enzyme Casitas b-lineage lymphoma proto-oncogene-b (Cbl-b) in the ubiquitin-proteasome pathway. Cbl-b-IN-2 can be used to modulate the immune system and diseases that are susceptible to immune system regulation. Cbl-b-IN-2 (Example 8) can also be used alone or in combination with one or more immune checkpoint inhibitors, anti-tumor agents, and radiation for cancer treatment.
