Exatecan (DX-8951) ist ein nicht-prodrug Camptothecin (CPT)-Derivat, das eine signifikante Topoisomerase-I-Hemmung zeigt [1]. Exatecan (DX-8951) hemmte Topoisomerase I mit einem IC50 von 2,2 µM (0,975 µg/ml) [2].
Exatecan (DX-8951) und SN-38 hemmten die Topoisomerase-Aktivität mit IC50-Werten von 0,82 bzw. 2,3 µg/ml, Topoisomerase I wurde aus SUIT-2-Zellen gewonnen [3]. Exatecan (DX-8951) (20 ng/ml) induzierte DNA-Fragmentierung. In einem Extrakt aus behandelten SUIT-2-Zellen waren etwa 60% der DNA bei einer Exatecan (DX-8951) Konzentration von 20 ng/ml fragmentiert. Exatecan (DX-8951) (0,05 µg/ml, 24 h) induzierte die spezifischen Merkmale der Apoptose, nämlich Chromatinkondensation, Kernfragmentierung und zytoplasmatische Vakuolisierung in SUIT-2-Zellen [3].
Exatecan (DX-8951) zeigte in vivo Aktivität gegen eine Vielzahl menschlicher Tumorxenografts bei nackten Mäusen, einschließlich Magen-, Pankreas-, Kolon-, Brust-, Eierstock- und Lungentumoren [4]. Exatecan (DX-8951) unterdrückte das Wachstum von menschlichem Magenadenokarzinom SC-6, das in nackte Mäuse transplantiert wurde. Als die Mäuse dreimal im Abstand von 4 Tagen i.v. mit Exatecan (DX-8951) behandelt wurden, wurde eine signifikante Hemmung des Tumorwachstums über einen breiten Dosisbereich (3,325 mg/kg bis 50 mg/kg Gesamtdosis) ohne toxischen Tod beobachtet. Seine antitumorale Aktivität war dosisabhängig, und bei der höchsten Dosis von 50 mg/kg betrug die Tumorwachstumshemmungsrate 92% [2].
