Fusaric Acid, ein von der Fusarium-Art produziertes Mykotoxin, hat vielfältige toxikologische Wirkungen auf Pflanzen und Tiere. [1]
In vitro aktiviert die Behandlung mit 104 µg/ml Fusaric Acid in humanen Hepatozellulären Karzinomzellen (HepG2) posttranslational p53 als Reaktion auf DNA-Schäden. [2] In einem In-vitro-Effizienztest wurde gezeigt, dass Fusaric Acid in HepG2-Zellen die p53-Promotormethylierung bei Behandlungen mit 25, 104 und 150 µg/ml Fusaric Acid bemerkenswert erhöhte; jedoch wurde bei der Behandlung mit 50 µg/ml Fusaric Acid die Promotormethylierung von p53 signifikant verringert. [1] In HepG2-Zellen erhöhte Fusaric Acid die Promotormethylierung von DNMT1 im Vergleich zur Kontrolle dramatisch; jedoch verringerte die Behandlung mit 25, 50 und 104 µg/ml Fusaric Acid die Promotormethylierung von DNMT3A und die Behandlung mit 150 µg/ml erhöhte die Promotormethylierung von DNMT3A. [3] Fusaric Acid hat eine Toxizität (24 h Inkubation; IC50 = 104 μg/ml) auf mitochondriale Leistung, zelluläre und mitochondriale Stressreaktionen, mitochondriale Biogenese und Marker des Zelltods. [4] Darüber hinaus zeigt Fusaric Acid eine Zytotoxizität gegenüber PBMCs mit einem IC50 von 240,8 μg/ml und Thp-1 mit einem IC50 von 107,7 μg/ml Zellen nach 24 h. [5]
In einem In-vivo-Experiment wurde gezeigt, dass die Behandlung mit 100 mg/kg Körpergewicht Fusaric Acid intraperitoneal 30 Minuten vor Beginn der Dunkelphase (Licht aus) bei Ratten das Niveau von Serotonin (5HT), 5-Hydroxyindolessigsäure (5HIAA), Tyrosin (TYRO) und Dopamin (DA) erhöhte und das Niveau von Noradrenalin (NEpi) verringerte.
