IRBP (1-20), human ist das 1-20 Aminosäurefragment des optischen Interrezeptor-Retinoid-bindenden Proteins (IRBP). IRBP ist ein 140 kDa Glykoprotein, das in der Photorezeptormatrix zwischen der neuronalen Retina und dem retinalen Pigmentepithel gefunden wird. Humane IRBP-Peptide 1-20 enthalten ein Haupt-Epitop des H-2b-Haplotyps. Die Immunität gegen IRBP (1-20) induziert eine T-Zell-vermittelte experimentelle autoimmune Uveoretinitis (EAU) [1,2].
Weibliche C57BL/6J-Mäuse, die durch IRBP (1-20), human induziert wurden, zeigten erfolgreich das EAU-Phänotyp [3,5]. Die Konsequenzen eines IRBP-Mangels auf das T-Zell-Repertoire wurden ebenfalls untersucht. IRBP+/+, IRBP+/- und IRBP-/- Mäuse auf dem C57BL/6-Hintergrund wurden mit IRBP oder einem pathogenen Epitop, dem IRBP (1-20), human Peptid im Adjuvans, immunisiert und auf Krankheitschwere und immunologische Reaktionen hin bewertet. C57BL/6 IRBP-/- Mäuse waren vollständig resistent gegen EAU und EAP und zeigten verstärkte immunologische Reaktionen auf IRBP und sein pathogenes Peptid 1-20 im Vergleich zu ihren IRBP+/+ Gegenstücken. IRBP-/- Mäuse zeigten eine veränderte IRBP-Epitoperkennung [4].
