Lometrexol (DDATHF)-Hydrat, ein Antipurin-Antifolat, kann die AktivitÄt der Glycinamid-Ribonukleotid-Formyltransferase (GARFT) hemmen, fÜhrt jedoch nicht zu nachweisbaren DNA-StrangbrÜchen. Lometrexolhydrat kann die De-novo-Purinsynthese weiter hemmen, was zu abnormaler Zellproliferation und Apoptose und sogar zum Stillstand des Zellzyklus fÜhrt. Lometrexol-Hydrat hat Antikrebs-AktivitÄt. Lometrexolhydrat ist auch ein starker Hemmer der menschlichen Serin-Hydroxymethyltransferase 1/2 (hSHMT1/2).

Lometrexol hydrate (DDATHF hydrate), an antipurine antifolate, can inhibit the activity of glycinamide ribonucleotide formyltransferase (GARFT) but do not induce detectable levels of DNA strand breaks. Lometrexol hydrate can further inhibit de novo purine synthesis, causing abnormal cell proliferation and apoptosis, even cell cycle arrest. Lometrexol hydrate has anticancer activity[1][2].
