Pemafibrate
Pemafibrat (K-877) ist ein oral verabreichter Peroxisom-Proliferator-aktivierter Rezeptor (PPAR)-α-Agonist zur Behandlung von Hyperlipidämie, EC50 auf Gal4hPPARα = 1 nM.
Pemafibrat (10 µM, 24 h) regulierte die Expression mehrerer Zielgene, die für Proteine kodieren, die am Kohlenhydrat- und Lipidstoffwechsel beteiligt sind, in primären menschlichen Hepatozyten und in der Leber von Mäusen. Pemafibrat (50 nM, 24 h) aktivierte die PPAR-α-Transkriptionsaktivität und war wirksamer als Fenofibrat und Pirinixsäure (Wy14643).
Pemafibrat (0,001% in MF-Diäten für 1 Woche) reduzierte signifikant den Triglycerid- und Gesamtcholesterinspiegel im Plasma, erhöhte den HDL-Cholesterinspiegel im Plasma, regulierte die Genexpression im Zusammenhang mit dem Triglycerid- und HDL-Cholesterinstoffwechsel in der Leber und regulierte die Genexpression des Cholesterin- und Triglyceridstoffwechsels im Dünndarm von Mäusen. Pemafibrat förderte auch den Cholesterin-Efflux und den umgekehrten Cholesterin-Transport, wirkte entzündungshemmend und verringerte atherosklerotische Läsionen. Pemafibrat war wirksamer als Fenofibrat bei der Unterdrückung des postprandialen Anstiegs der Chylomikronen und der Anhäufung von Chylomikronenresten, wodurch es die postprandiale Hypertriglyceridämie abschwächte.
