Phos-tag is an innovative phosphate-binding tag and functional molecule that can specifically bind to phosphate ions under neutral pH (physiological pH) conditions and is often used to analyze the phosphorylation level of proteins. This bidentate metal complex (1,3-bis[bis(pyridin-2-ylmethyl) amino]propan-2-olato dizinc(II) complex) exhibits remarkable selectivity, allowing stable binding to diphenyl phosphate dianions (PH-OPO32-) in neutral pH aqueous solutions, with a Kd value of 25 nM (in the presence of Zn2+). Phos-tag provides a unique electrophoresis analysis technique known as manganese (II)-Phos-tag SDS-PAGE, which enables simultaneous analysis of protein phosphorylation and non-phosphorylated forms.

Separation Principle:

  1. Phosphorylated proteins in electrophoresis capture two divalent metal ions.
  2. The higher the level of phosphorylation, the slower the electrophoretic mobility.
  3. Phos-tag separation separation is based on the degree of phosphorylation (even if the number of phosphorylation sites is the same, separation can occur as long as their positions differ, ).
