HG-9-91-01

HG-9-91-01 ist ein salzinduzierbarer Kinase (SIK) Inhibitor, der SIK1, SIK2 und SIK3 mit IC50-Werten von 0,92nM, 6,5nM und 19,4nM hemmt [1].

HG-9-91-01 hemmt die SIKs stark und beeinflusst kein anderes Mitglied der AMPK-verwandten Kinase-Unterfamilie. HG-9-91-01 erhöhte die LPS-stimulierte IL-10-Produktion und unterdrückte die Sekretion proinflammatorischer Zytokine [2]. Eine Vorbehandlung von RAW264.7-Zellen mit 0,5 µM HG-9-91-01 für 30 Minuten vor der RANKL-Stimulation führte zu einer Reduktion der Bildung mehrkerniger Zellen und der TRAP-Färbung und reduzierte die mRNA der Osteoklastendifferenzierungsmarker in einer konzentrationsabhängigen Weise signifikant. Die Vorbehandlung mit 0,5 µM HG-9-91-01 verringerte die Bildung von Resorptionslakunen im Vergleich zu RANKL allein signifikant [3].

HG-9-91-01 (3, 10 oder 30 mg/kg) erhöhte die Expression von IL-10, während es die Spiegel von IL-12 und TNF-α im Kolongewebe von TNBS-Mäusen verringerte [4].
