Theaflavin (TF) est un dérivé distinctif des benzotropolones, extrait pour la première fois du thé noir, et possède des effets antioxydants grâce à ses fonctions de piégeage des radicaux libres et de chélation des métaux[1,2]. De plus, la théaflavine est un inhibiteur naturel efficace contre la neuraminidase du virus de la grippe A (H1N1) [3].
La théaflavine (20 et 50 µg/ml; 2 h) a inhibé les lésions oxydatives induites par le monohydrate d'oxalate de calcium (COM) dans les cellules épithéliales tubulaires rénales HK-2 [4]. Les cellules endothéliales veineuses ombilicales humaines (HUVECs) exposées à la théaflavine (0-200 µM; 24 h) peuvent atténuer l'apoptose et la dysfonction cellulaire induites par le tert-butyl hydroperoxyde (TBHP) [5]. La théaflavine (0-160 µM; 72 h) a inhibé la prolifération des cellules CNE2, favorisé l'apoptose, augmenté l'expression des caspases 3, 9 et Bax, et diminué le niveau de Bcl-2 [6].
La théaflavine (20, 50, 100 mg/kg; 14 jours; par voie orale) a atténué les lésions oxydatives et les dépôts cristallins induits par la néphrocalcinose à CaOx chez les souris [4]. Dans un modèle murin de lésion rénale par ischémie/reperfusion (I/R), un prétraitement à la théaflavine (20/50/80 mg/kg; 3 jours; par voie orale) a significativement inhibé les lésions rénales et l'apoptose induites par l'I/R. Elle a amélioré la dysfonction mitochondriale en réduisant les dommages mitochondriaux et en favorisant le potentiel de membrane mitochondriale [7].
