4-メトキシトリチル(Mmt)は、非常に酸に敏感なシステイン保護基であり、0.5〜1.0%TFAとスカベンジャー(TESなど)の存在下で選択的に除去することができます。t-ブチルタイプやS-Trtの保護基はこれらの条件下でも不変です。ヨウ素を添加することでクレージングと環化を同時に行うことができます。システイン含有ペプチドの固相合成およびラベリンググループ導入のために、さらにチオール機能を選択的に脱保護するために使用されることがあります。Rijkersらは、p-メトキシトリチル保護を用いてレジン上でS-パルミトイル化ペプチドを得るために使用しました。Cys(Mmt)の高い酸性耐性はRobertsらによって硫エーテル結合した環状ペプチドライブラリーを得るために利用されました。Arendtらは、パーフォーミック酸でThiol部位を選択的脱ブロッキングおよび酸化してシステイン酸含有ペプチドの合成にCys(Mmt)を使用しました。MmtはHOBtに完全に安定しています。
