Aflatoxin B1は、アスペルギルス菌によって生産される発癌性のマイコトキシンの一種であり、人間や動物において肝細胞癌(HCC)の発症を常に引き起こす。Aflatoxin B1の毒性は急性毒性、胎児異常性、変異原性、発癌性を含む。また、Aflatoxin B1によって引き起こされるDNA付加、炎症、酸化ストレスもがんの発症に関与する。
体外実験では、AFB1はHek293細胞の細胞生存率を32.60 µMのIC50で抑制することが示されている。体外実験では、0、10、50、100 µMのAFB1で卵母細胞の成熟が行われた。50および100 µMのAFB1濃度では、メタフェーズII段階に到達する卵母細胞の数が減少し、卵母細胞の細胞内GSHレベルが低下した。また、成熟した卵母細胞における細胞内ROS産生も最高レベルに達した。
体内実験では、1日5 mg/kgのAflatoxin B1を雛鶏に腹腔内投与すると、ASTおよびALTレベル(肝障害の指標)が著しく上昇した。また、Aflatoxin B1による亜急性または慢性曝露後の動物のパフォーマンスの低下が示され、LD50値はヒヨコで0.3 mg/kg体重、マウスで9.0 mg/kg体重、雌ラットで17.9 mg/kg体重まで変動した。妊娠中のマウスに対して高用量の20 mg/kgのアフラトキシンB1を腹腔内投与したところ、血中で15分後にピーク吸収が観察され、経口摂取後は30分後にピークが観察された。これは、アフラトキシンが近位腸からの吸収が非常に迅速であることを示唆している。
体内実験では、高用量の75 µg/kgのアフラトキシンB1では、糞便中の短鎖脂肪酸(SCFA)レベルが70%以上減少した。さらに、25 µg/kgのアフラトキシンB1を経口投与されたラットでは、糞便中の胆汁酸(2.18倍)、リノール酸(cis-9、cis-12-18:2)(11.3倍)、ペンタデカン酸(15:0)(3.68倍)、ピルビン酸(4.56倍)、3-フェニル乳酸(3.74倍)が顕著に増加した。
