SB 290157 (trifluoroacetate salt)

SB 290157 (trifluoroacetate salt)は、選択的な非ペプチドC3aR拮抗剤として使用される。SB 290157は、ヒトC3aRを発現するラット好塩基性白血病細胞に対して選択的な拮抗剤であり（IC50は200 nM）、この拮抗作用はヒトC3aR（IC50は12 nM）に限定されず、マウスおよびモルモットC3aRを発現する細胞のC3a誘導Ca2+動員も抑制した（それぞれIC50は7および30 nM）。

SB 290157（トリフルオロ酢酸塩）（0.1 µM）は、SHRラットのVSMCにおいてSM22α mRNAの量を有意に増加させ、オステオポンチンmRNAの量を減少させたが、WKYラットの細胞ではこれらの変化は観察されなかった。C3aの過剰発現はヒトポドサイト細胞株におけるビンキュリンの分布を著しく変化させたが、1 µMのSB 290157によるC3aR阻害はこの変化を阻止した。SB 290157（0.1-10 µM）は、10 nMのC3aによって誘導されるC3aRの内部化を濃度依存的に抑制した。1 µMを超える濃度のSB 290157の存在下では、C3aによって誘導されるC3aRの内部化が約50%減少した。

SB 290157（トリフルオロ酢酸塩）（1 mg/kg）の治療は、リポポリサッカライド（LPS）誘導マウスにおけるミクログリアの活性化および認知障害を減少させた。SB 290157（トリフルオロ酢酸塩）は、LPS誘導の海馬神経炎症および抑制性シナプス関連タンパク質の喪失を軽減し、マウスの認知機能を改善した。SB 290157（トリフルオロ酢酸塩）は、抗OVA pAb誘導関節炎の抑制に関与し、30 mg/kgの濃度でSB 290157を注射すると、3時間後に関節の腫れを減少させ、約50%の関節腫れの抑制を達成した。
