Description of 7-keto Cholesterol

7-keto Cholesterol은 혈관 질환과 밀접한 관련이 있는 주요 옥시스테롤이다. 이 물질은 동맥 경화증 플라크에 널리 존재하고 콜레스테롤보다 강한 동맥 경화 유발 효과를 가지고 있다. 7-keto Cholesterol은 담즙산과 콜레스테롤 합성에 관여하는 제한속도 효소를 억제할 수 있다. 예를 들어 콜레스테롤 7-hydroxylase와 HMG-CoA 환원 효소 등이 있다[2]. 게다가 7-keto Cholesterol은 염증을 유발하는 작용을 가지고 있다[3].

체외 실험에서 7-keto Cholesterol(50μM)로 인간 제대 정맥 내피 세포(HUVECs)를 18시간 처리한 결과 접착 분자의 발현(특히 E-셀렉틴)이 상향 조절되었고 IL-8 mRNA의 발현이 증가했고 NF-κB의 활성이 증가했다[4]. 7-keto Cholesterol(2.5-40μg/mL)로 인간 미세혈관 내피 세포(HMVECs)를 처리한 결과 농도 의존적으로 세포 생존력이 억제되었고 시스테인 아스파라긴-3/7, -8 및 -12가 활성화되었다[5]. 7-keto Cholesterol(5μg/mL)로 혈관 평활근 세포(VSMCs)를 24시간 처리한 결과 인터루킨-6(IL-6)의 발현이 현저히 증가했다[6].

체내 실험에서 7-keto Cholesterol(0.1%)을 허혈-재관류(IR) 손상을 받은 쥐의 사료에 3주 동안 첨가한 결과 쥐 혈장 중 7-keto Cholesterol의 농도가 증가했고 IR 후 심근 경색의 크기가 증가시켰다[7].

제조 방법

HUVECs는 50μM 콜레스테롤 또는 7-keto Cholesterol로 18시간 전처리한 후 0.1ng/mL TNF-α로 추가 4시간 처리했습니다. RIPA 분해 버퍼는 프로테아제와 인산효소 억제제를 사용하고 HUVECs로부터 신선한 총 단백질 추출물을 분리했습니다.

응용 분야

7-keto Cholesterol은HUVECs에서 접착 분자의 발현을 증가시켰습니다. 특히 E-selectin에서 두드러졌습니다. 7-keto Cholesterol은 또한 IL-8 mRNA의 발현을 증가시켰지만 MCP-1의 발현에는 영향을 미치지 않았습니다.
