Biotin-XX Tyramide Reagent는 에이징트가 중개하는 타이라미드 신호 증폭(TSA)을 위한 비오틴 유도체이다. Biotin-XX Tyramide Reagent는 호세라드시스페로시드아제(HRP)를 이용하여 고정된 에이징트 옆에 비오틴 레이블을 공유적으로 쌓아 넣다. 쌓인 레이블은 브라이트 필드나 플루오레상현미경으로 영상을 얻기 위해 스트레프타비딘 결합물로 감지할 수 있다. 레이블링 반응은 빠르게 진행된다(10분 미만)[1]. Biotin-XX Tyramide Reagent는 면역조직화학의 민감도를 높이는데 사용할 수 있다[2].

Protocol of Biotin-XX Tyramide Reagent

1.타이라미드 신호 증폭 (TSA) 준비

1.타이라미드 신호 증폭 (TSA) 준비: 문헌[1]의 방법을 참고하여 TSA를 다음과 같이 준비했습니다: Biotin-XX Tyramide (품번: GC12033), DAPI 포함 항색변형 마운팅 매디엄 (품번: GC26283), HRP-결합 스트레프타비딘 (품번: GC26343), 스트레프타비딘-FITC (품번: GC19886), TNT 버퍼 (pH 7.5) (0.1 M 트리스-HCl, 0.15 M NaCl, 0.05% 트윈 20을 포함), TNB 버퍼 (TNT 버퍼 + 0.5% BSA).

2.HRP 스트레프타비딘 결합:

① TNB 버퍼로 비오틴 표시된 헤라 세포를 30분 동안 차단합니다.

② HRP 스트레프타비딘을 TNB 버퍼에서 1:1000으로 희석하여 헤라 세포에 추가하고 30분 동안 발효시킵니다.

③ TNT 버퍼로 세포를 세 번 씻어냅니다. (TNB 버퍼의 BSA는 차단제로 사용됩니다).

3.TSA 증폭:

① 증폭 작업 용액에서 헤라 세포를 10분 동안 차단합니다 (증폭 작업 용액에는 0.1 M 보라트 버퍼 (pH 8.5)에 1 μg/mL Biotin-XX Tyramide Reagent (BXXP)와 1 mM H2O2가 녹아 있습니다).

② TNT 버퍼로 세 번 씻어냅니다.

4. 스트레프타비딘-FITC 결합:

① TNB 버퍼로 비오틴 표시된 헤라 세포를 20분 동안 차단합니다.

② TNB 버퍼에 1.0 μg/mL 스트레프타비딘-FITC를 추가한 후 헤라 세포를 추가하고 30분 동안 발효시킵니다.

③ TNT 버퍼로 세포를 세 번 씻어냅니다.

5. 형광 현미경:

①현미경 슬라이드에 DAPI를 포함한 항색변형 장착 매체 (10 μL)을 추가합니다.

② 24웰 판에서 헤라 세포가 있는 유리 커버리프를 뾰족한 핀셋으로 제거하고 DAPI가 포함된 항색변형 마운팅 매디엄 위에 추가합니다 (세포 면을 아래로).

③ 자외선 발생과 방출 광 조건이 495nm/525nm (녹색)인 상황에서 형광 현미경으로 관찰합니다.
