Description of C29

C29는 Toll-like receptor 2 (TLR2) 억제제이다.C29는 인체 HEK-TLR2와 THP-1세포에서는 TLR2/1 및 TLR2/6경로를 억제하지만, 쥐 대식세포에서는 TLR2/1 경로만을 억제한다.C29는 다른TLR흥분제와 TNF-α 유도 신호전도를 억제하지 못한다. BB 루프 포켓 잔류물는 TLR2/1이TLR2/6이 아니지만 TLR2/6은 아닌 신호로, 서로 다른 역할을 암시했다.

C29 TLR2/1-mediated 억제 NF-κ B의 활성화를 억제하고 (IC50. 87 µ M), TLR2/6 억제와 heterodimer IC50 23 µ M를 억제한다.C29와 o-vanillin은TLR2 TIR 영역을 특정적으로 BB 루프 포켓을 타겟으로 하여 그 기능 및/또는 위치를 변화시킴으로써 기능할 수 있다.

 

B e antigen (HBeAg)을 투여하기 전에 C29로 전처리하여 쥐에게 TLR-2의 효과를 확인한 결과 IL-6, TNF-α, CCL-2의 발현은 감소하였으나 IL-10은 간에서 상위되었다.C29는 지질의 축적에 영향을 미치지 않았으나 EPS는 지방 형성에 대한 억제작용이 현저하게 감소되었다.그 결과, 초기 (4일)에 C29에 의해 TLR2와 88 (MyD88)의 발현이 억제되었을 때EPS는 AMPK 신호 경로의 활성화를 억제하였으며, 이는 EPS 가 TLR2와 상호작용하여AMPK신호 경로의 활성화를 억제함을 의미한다 .

제조 방법

C29는 DMSO에 녹여 50mM의 보관용 솔루션으로 만들었습니다. 세포 재배에서 최종 DMSO 농도는 0.2% (v/v) 미만이었습니다. 세포는 먼저 C29와 1시간 동안 배양한 후에 해당 TLR(Toll 유사 수용체) 가동제로 자극되었습니다.

반응 조건

0.01µM, 0.1µM, 1µM, 10µM 100µM ,1 시간 동안
응용 분야C29는 TLR2/1 또는 TLR2/6에 대한 억제 작용을 보였습니다 (IC50는 각각 21µM과 23µM).

제조 방법

C29는 용액에 녹이고 희석되었습니다 (10% DMSO/40% PEG300/5% 튜브인-80/45% 식염수), 그리고 HBeAg 처리 1시간 전에 복강내 주사로 투여되었습니다 (1.3µmol/g), 용액 그룹과 동일한 용량의 용제가 사용되었습니다.

응용 분야체내 TLR-2의 역할을 확인하기 위해 쥐는 HBeAg 투여하기 전에 C29로 사전 처리되었습니다. IL-6, TNF-α 및 CCL-2의 표현은 현저하게 완화되었지만 간에서 IL-10은 상향 조절되었습니다.
