BRET 검사에서 Coelenterazine 400a에 대한 실험 프로토콜

1. 원액의 준비: 100% 에탄올이나 메탄올에 Coelenterazine 400a를 녹여 농도 1mM의 원액을 준비합니다. 원액을 나눠 -20°C나 -80°C의 어두운 곳에 보관합니다.

2. 검체의 준비: 적절한 매체에서 세포를 재배하여 70-80%의 혼잡도에 이르게 합니다. 적합한 전염제를 사용하여 Renilla luciferase와 형광 수용체 플라스미드를 함께 전염합니다. 전염 후 충분한 단백질 표현을 보장하기 위해 24-48시간 동안 세포를 계속 재배합니다.

3. 기판의 준비 용액: 사용하기 전에 Coelenterazine 400a 원액을 세포 재배 매체로 100μM 농도로 희석합니다.

4.BRET 검사: 전염된 세포를 세척하고 인산완충 식염수 (PBS)에 소생시킨 후 96웰의 화이트 마이크로 플레이트의 각 컵에 90μL의 세포 소생된 용액을 추가합니다. 각 컵에 10μL의 기질 용액을 추가하여 최종 농도를 10μM로 만듭니다.
