Z-DEVD-FMK

Z-DEVD-FMK은 특이성이 있고 불가역적인 반고신-아스파라긴 프로틴아제 3(caspase-3) 억제제로서 IC50가 18μM이다. Z-DEVD-FMK은 일반적으로 세포의 조절된 사멸과 신경 보호를 연구하는 데 사용된다^[2].

체외 실험에서 Z-DEVD-FMK(50μM)은 심장 근세포를 24시간 처리하여 세라미드 유도된 세포 사멸을 현저하게 감소시켰으며, 세포 생존률을 81%까지 높였다^[3]. Z-DEVD-FMK(15μM)은 신장 근처 소관 세포를 치료할 때 caspase-3 활동을 효과적으로 감소시킬 수 있지만, 세포의 조절된 사멸을 감소시키는 효과는 범-caspase 억제제에 비해 좋지 않는다^[4].

체내 실험에서 Z-DEVD-FMK(160ng)은 뇌수액관 주입을 통해 외상성 뇌손상(TBI)을 가진 쥐를 치료하여 중추신경계 손상 후의 병변 부피를 감소시키고 운동 및 인지 기능을 개선시켰다^[2]. Z-DEVD-FMK(20mg/kg)은 복강 주입을 통해 당뇨병 쥐를 8주간 치료하여 쥐의 단백뇨, 신장 기능 및 신장 소관간질 섬유화를 개선하고 혈청 흰 단백질 수준의 감소를 늦추었다^[5]. Z-DEVD-FMK(0.2mg/kg)은 눈동자에 주사하여 토끼의 시신경 손상을 치료하여 망막 신경절로 세포의 조절된 사멸을 현저하게 감소시키고 시신경 기능의 회복을 촉진시켰다^[6].
