Description of Zinpyr-1

Zinpyr-1 (Zinc (Zn)-fluorophore 1, ZP1)은 칼슘 검출을 위한 형광 프로브로 밝은 형광을 방출하고 칼슘 표지에 널리 사용되었다 (Ex/Em: 515nm/531nm) (ε = 79,000/M/cm; pKa = 8.4; Φ = 0.38). Zinpyr-1은 표형광, 공초점 그리고 2 광자 현미경과 유세포 분석기 측정과 호환되고 살아있는 세포와 고정된 세포의 응용에 사용할 수 있다. Zinpyr-1은 동물, 식물, 세균의 체외, 체내 재료와 모델에서 널리 사용되고 있고 예를 들어 인간 혈청, 아랍리피드 뿌리, 쥐 그리고 대장균 등에서 사용된다[1]. Zinpyr-1은 형광 유도체 리포터와 금속 결합 유닛으로 구성되어 있고 Zinpyr-1이 Zn²⁺을 결합하면서 형광 신호를 방출할 수 있다[2].

Protocol of Zinpyr-1

원액 준비:

1.Zinpyr-1을 DMSO에 10 mM로 녹인 후 -20°C에서 보관하고 반복적인 냉동-해동을 피하도록 합니다.

2.적절한 용매(예: Zn²⁺이 없는 배지)로 Zinpyr-1 작업 용액을 준비합니다. Zinpyr-1 작업용액의 농도는 실험 요구에 따라 결정됩니다.

유세포 분석기를 사용한 Zinpyr-1로 세포 내 Zn²⁺ 정량화[1]:

1.샘플을 준비합니다(정지된 세포를 소생시키고 부착세포는 트립신-EDTA로 소화합니다).

2.HEPES 버퍼링된 “zinc free” RPMI 배지를 준비합니다(1 mM EDTA를 포함하는 RPMI 배지).

3.HEPES 버퍼링된 “zinc free” RPMI 배지에서 Zinpyr-1(20 µM)과 함께 37°C, 5% CO₂에서 30분 동안 세포를 배양합니다.

4.1 mM EDTA가 포함된 “zinc free” RPMI 배지(1 mL)로 세포를 3번 세척합니다.

5.상청액을 버리고 세포를 500 µL PBS로 소생시킵니다.

6. 어두운 곳에서 항체(이 경우엔 R-phycoerythrin-conjugated mouse anti-human monoclonal antibody와 PE-Cy5-conjugated mouse anti-human monoclonal antibody)와 함께 4°C에서 20분 동안 세포를 배양합니다. 7. 1 mM EDTA가 포함된 “zinc free” RPMI 배지(1 mL)로 세포를 2번 세척합니다. 8. 유세포 분석기로 측정합니다.

Zinpyr-1을 사용한 살아있는 세포 내 Zn²⁺ 가시화:

1.HEPES 버퍼링된 “zinc-free” RPMI 배지를 준비합니다(1 mM EDTA를 포함하는 RPMI 배지).

2.HEPES 버퍼링된 “zinc-free” RPMI 배지에서 Zinpyr-1(20 µM)과 함께 37°C, 5% CO₂에서 30분 동안 세포를 배양합니다.

3. 1 mM EDTA가 포함된 “zinc-free” RPMI 배지(1 mL)로 세포를 3번 세척합니다. 4. 형광 현미경에서 결과를 확인합니다(Ex/Em: 515nm/531nm).
