Cy3-dCTP can be used for PCR by Taq polymerase, Nick Translation by DNAse I/DNA polymerase I, cDNA labeling and 3' end labeling for direct enzymatic labeling of DNA/cDNA. Cy3-dCTP can replace CTP in reactions as a substrate for T4 and Taq DNA polymerases, E. coli DNA polymerase (holoenzyme and Klenow fragment), reverse transcriptase (from AMV and M-MuLV) and terminal transferase. Synthetic dye-labeled DNA/cDNA probes can be used to identify specific sequences by in situ hybridization, microarray or blotting techniques. Cy3-dCTP can also be used for multicolor fluorescent labeling. An optimized linker attached to the C5 position of uridine ensures optimized substrate properties and labeling efficiency. Recommended Cy3-dCTP/dCTP ratio for PCR and Nick Translation: 30-50% Cy3-dCTP/50% dCTP.
